Gene therapy using adeno-associated virus (AAV) as a vector has emerged as a novel therapeutic modality that has the potential to cure many diseases. Currently, there are over 140 in clinical trials stage. By 2025, FDA predict that 10 to 20 cell and gene therapy products will be approved a year based on an assessment of the current pipeline and the clinical success rates of these products.

Currently, ddPCR (Droplet Digital PCR) or qPCR (quantitative) are quantitative methods heavily used in the AAV discovery and development process. These nucleic acid based method can provide accurate AAV genome titer. However, the disadvantages of the methods include:

— Rely on costly instrument

— Require special designed laboratory and specialized personal.

— Time consuming.

A rapid, instrument-free and simple method to check AAV titer is needed to facilitate AAV discovery and development process.

Here, we developed AAV lateral flow based strip for the semi-quantitative detection of AAV capsid titer in just few minutes. It can be used for crude lysate or purified material to estimate and compare AAV titer. This AAV strip can be adapted to monitor and in-process control AAV production and purification process. The unique features of our AAV strip include:

— Rapid: few minutes to readout

— Easy to use: Without special training and no expensive equipment needed

— Serotype-specific: serotype-specific AAV antibodies were developed to recognize unique serotype AAV
